Abstract:
This chapter has two sections. The first includes an overview of the history of ancient DNA research. New approaches and modern analysis protocols are explained, with much reference to next-generation sequencing and the current perspectives in the study of prehistoric populations. The proper methods for handling and sampling the osteological material are discussed, and special attention is paid to the shortcomings of the new methodology, such as contamination, sample quantity, and genetic material conservation. The second section briefly discusses the uses of paleogenetics in anthropology, molecular biology, contemporary population genetics, and forensics. The chapter ends providing examples of research on gene mapping in modern and prehistoric populations, the geographic distribution of human genetic diversity, and the reconstruction of their biological histories.
