Abstract:
The chapter includes four exercises. The first aims to familiarise students with chromatographic and spectrophotometric methods for the isolation, qualitative and quantitative analysis of biomolecules, using photosynthetic pigments (chlorophylls and carotenoids) as a model. The second detects and quantitatively measures photosynthetic electron flow and oxygen release in isolated chloroplasts. The third analyses the role of starch as a storage material, and the dependence of its biosynthesis and catabolism on the presence of light, indicating the competition between photosynthesis and respiration in the direction of metabolism. Finally, the fourth compares the leaf anatomy (by microscopic observation of fresh sections) and the specificities in gas exchange between two photosynthetic variants (C3 and C4). The aim is to understand the adaptive advantages of each variant and to familiarise the methods for measuring photosynthetic CO2 assimilation.
