Cytogenetics is the scientific field of genetics that studies the number and structure of human chromosomes. Classical cytogenetic analysis is used in clinical practice for the diagnosis of syndromes, the investigation of mosaicism cases, infertility, recurrent miscarriages, short stature, ambiguous genitalia, and more. In many cases of neoplasms, chromosomal rearrangements carry both diagnostic and prognostic significance. By observing chromosomes under a microscope, cytogenetics can detect rearrangements of approximately 5–10 Mb in size. For smaller genetic rearrangements, techniques with higher resolution can be applied, such as molecular cytogenetic methods (e.g., Fluorescent In Situ Hybridization - FISH, multicolour FISH - mFISH, array-CGH, etc.). Comparative Genomic Hybridization microarray technology (array-CGH), also known as "Molecular Karyotyping", bridges the diagnostic gap between classical cytogenetics and molecular genetics. It offers 10 to 1000 times greater resolution, although it cannot detect qualitative changes such as inversions and balanced translocations. It is a powerful methodological tool for studying patients with unexplained intellectual disability, multiple congenital anomalies, and more. It largely constitutes an automated molecular method for analyzing Copy Number Variations (CNVs) in DNA.
