Abstract: |
Chapter 2 presents the chemical composition of biological membranes. In detail, in the subchapter dealing with membrane lipids, chemical formulae, representative chemical reactions and physicochemical properties and methods of analysis are provided. A special section is devoted to lipid polymorphism. The physicochemical background of phospholipids’ super-molecular assembly and organisation in aqueous solutions, concepts such as Critical Micellar Concentration (CMC), as well as relevant study techniques are presented. In the subchapter devoted to membrane proteins, methods of isolation and protein techniques, including solubilisation using specific detergents, chromatographic methods of purification, the concept of the hydrophobic-hydrophilic constant and reconstitution techniques are presented in detail. Next, we present characteristic examples of membrane proteins with particular biological, physiological or structural importance. Modern biotechnological applications are described, such as the utilisation of liposomes in drug delivery, enzymic studies, as well as in environmental applications, as biosensors. Finally, a special subchapter is devoted to the study of membrane proteins and the management of high throughput data with bioinformatics tools.
