Abstract:
The purpose of the exercise is to introduce the trainees to gel electrophoresis, one of the most basic techniques for the separation and isolation of nucleic acids and proteins. The technique is based on the mobility of the molecules according to their size as they move through a gel while an electric field is applied to the ends of the gel. The introductory part of the exercise introduces the principles of electrophoresis and describes the techniques of DNA and protein electrophoresis on agarose and polyacrylamide gels, respectively, as well as some variations and applications of these techniques. In the practical part, students are trained in the technique of DNA electrophoresis on agarose gel, using different DNA mixtures, in order to understand how the shape of the molecules affects their movement and thus their separation during electrophoresis. The exercise allows trainees to understand the physical properties (charge, size) of DNA and proteins, and the possibilities for research provided by the ability to separate and isolate the two classes of macromolecules.
