Abstract:
The purpose of the exercise is to familiarize the trainees with the fertilization of the egg, the first divisions of the zygote and the onset of cellular differentiation in the early stages of embryogenesis. For this purpose, gametes and zygotes of the sea urchin are used because of their many advantages in the experimental handling (ease of obtaining gametic material, ease of in vitro fertilization of the oocyte, large and isolecithin oocyte, ease of microscopic observation of the first divisions). The exercise gives the opportunity to observe 'live' under the microscope the fertilization of the oocyte by the spermatozoa and (with appropriate advance preparation) the stages of two, four, eight, sixteen, thirty-two and sixty-four cells (morula to blastula stage). It also allows to discuss the phenomena of stem cell totipotency and cell differentiation.
