Abstract:
The aim of the exercise is to understand the mechanisms of nuclear division in the animal and plant cell. The introductory part introduces mitosis and meiosis as part of the cell cycle and describes the structure, morphology and movement of chromosomes. This is followed by a discussion of the stages of mitosis and the morphological modifications visible under the microscope. The practical part describes the experimental procedure for the observation of mitotic chromosomes in permanent microscope slides and in temporary ones from fresh onion root tips suitably treated and stained with Feulgen dye. In addition, information is provided for the study of the karyotype of human and representative plant species, for the observation of polytene chromosomes (large chromosomes which have thousands of DNA strands), and the reproductive processes in the algae Spirogyra (monogenic-amphigenic reproduction) observed microscopically in the context of the exercise. At the end of the exercise the student is asked to answer self-assessment questions.
