Abstract:
Deciphering the functionality of the cell was made possible by a combination of observation of cellular structures and biochemical analysis. Exercise 1 of this laboratory guide is about introducing the student to the modern compound microscope, in order to learn how it works and how to use it. In the introductory section, the basic principles of microscopy are discussed and the two types of microscopes (photon, electron) and their main variants (phase contrast microscope, fluorescence microscope, etc.) are presented. The parts of the modern compound microscope (optical and mechanical) are then described and a detailed description of the instrument's operations is given. Finally, instructions are given on how to prepare temporary microscope slides and methods of staining biological materials, and advice on how best to observe them. The student is prepared for laboratory practice in the observation of microorganisms, cells and tissues.
