Abstract:
The purpose of the exercise is to understand the properties of nucleic acids and to isolate deoxyribonucleic acid (DNA) from plant cells. The student will be trained in a simple procedure to extract DNA from strawberries (which, thanks to their octaploid genome allow the extraction of a large amount of genetic material) using everyday materials. In the introductory part of the exercise, the structure of DNA and the way it is coiled into chromatin is briefly described, and the methodology for estimating the concentrations and purity of nucleic acids in a solution is analyzed, specifically the widely used method of spectrophotometric analysis (measurement of the absorbance in ultraviolet light). The practical part gives a detailed description of the individual steps of the extraction procedure with the help of videos and animated diagrams (cell lysis - membrane dissolution, DNA precipitation, etc.), as well as the DNA quantification procedure.
